Study: Immunotherapy Efficacy Can Be Boosted By Ultrasound-Guided Microbubble

Ultrasound-guided cancer immunotherapy platform developed by the University of Texas MD Anderson Cancer Center promotes antitumor immunity and increases therapeutic efficacy. The research published in Nature Nanotechnology proposed a Micro-bubble assisted Ultrasound-guided Immunotherapy of Cancer (MUSIC) approach that consists of nano complexes combined with microbubble to effectively deliver cyclic guanosine monophosphate-adenosine monophosphate (cGAMP) to stimulate type 1 interferon essential for priming tumor-specific T cells. 

In the preclinical study, the MUSIC approach had demonstrated a 60% tumor eradication rate in breast cancer models in the monotherapy module. When combined with an anti-PD-1 antibody, the therapy improved antitumor response with minimal toxicity with decreased disease4 progression. The combined therapy also showed a superior survival benefit by increasing the median survival rate to 76% in comparison to monotherapy.

Wen Jiang, M.D., Ph.D., assistant professor of Radiation Oncology stated that the MUSIC approach provides a new opportunity to develop image-guided immunotherapy by using biomaterials to ensure efficiently, targeted, and robust immune activation to produce effective antitumor activity with minimal toxicity. He also added that the approach can be used for targeted delivery of other immune-stimulating agents such as Vaccines, mRNAs, gene therapy, etc.
